Description of Research:
Our investigations are focused on virulence pathways of bacterial pathogens, especially on the role of bacterial enzymes and proteins, among others proteases and peptidyloarginine deiminase, in the deregulation of a number of physiological pathways and evasion of host immunity. Our studies centre on Staphylococcus aureus and periodontal pathogens, including Porphyromonas gingivalis, Prevotella intermedia and Tannerella forsythia. The present studies are also aimed at elucidation of a novel protein secretion system (PerioGate) used by periodontopathogens to secrete they virulence factors, including proteinases.
Roles within RAPID:
Structural characterisation of PADs and other proteins of therapeutic interest
Shared PD/RA auto-antigens & tolerance breakdown due to NETs/citrullination

Description of Research:
Murine periodontal disease models will be employed to assess if P. gingivalis infection exacerbates RA. The potential influence of oral inflammation on systemic autoimmunity and inflammation will be investigated using transgenic animals and bacteria with molecular and cellular analysis. In addition, comparison of P. gingivalis W83 or PPAD deficient mutant will indicate whether PPAD modulates the autoimmune response.

Post-translational Modifications (PTMs) of proteins and their role in RA/PD
